Department of Biodiversity Protection is the only department belonged to the Institute situated in Popielno on a peninsula, in the reserve and in the Natura 2000 area (http://popielno.pl). There are equipped laboratories in the Department: biochemical, in vitro and molecular biology. Such a location provides isolation from the city noise, contact with nature, but at the same time, access to the nearest city (Ruciane-Nida), located about 20 km away, does not take more than 15-20 minutes.

Expectations:

  • directing the pursuit of research interests,
  • applying scientific and research projects and publication of the results in articles with IF,
  • developing methodological skills in biochemical, microbiological and molecular biology analysis,
  • communicativeness and good organization of work.

Scientific research and other tasks in which the Candidate would participate:

  • studies concerning microbiota and immunological parameters in the organism physiology of free-living animals,
  • research related to the genetic diversity of free-living animals using transcriptomic and proteomic methods,
  • research related to the preservation of gametes, gamete and embryo banking as well as breeding biotechnologies that can be implemented in free-living animals,
  • searching for breeding markers used in the development of effective techniques to reduce or intensify reproductive processes,
  • development of non-invasive hormonal monitoring in relation to reproduction, behavior, etc. in free-living animals.
  • influence of environmental factors, such as feeding base, antroporession on homeostasis of selected animal species,
  • work with live domestic animals and collection of research material from domestic and free-living animals, vividly and post mortem.

Candidate qualification requirements:

  • knowledge of animal physiology,
  • PhD degree in agricultural, biological, veterinary or related sciences,
  • good, documented by own research (in the form of PhD thesis, publications from the Philadelphia list and presentation of results at scientific conferences) knowledge of the subject of animal biology and behavior,
  • experience in the application of molecular biology methods and microscopic techniques and computer analysis of results, especially ELISA, Real Time PCR,
  • research internship outside the candidate’s home country,
  • participation in research projects,
  • participation in scientific conferences,
  • very good oral and written English skills, knowledge of another foreign language will be an advantage,
  • practical ability to drive a passenger car (category B, at least 2 years),
  • courses, training and practical skills related to animal work will be an advantage,
  • references of your experience in scientific research and analytical work will be an advantage.

The candidate will participate in the following research tasks:

  • isolation and culture of spermiophages from turkey semen,
  • immunofluorescence detection of the SSc5D in turkey semen spermiophages,
  • investigation of changes in spermiophage population (M1, M2 and SSc5D+) in relation to turkey semen quality,
  • analysis of the proteomic signature of activated spermiophages from turkey semen in relation to semen quality.

Conditions of work:

Requirements:

  1. PhD in biotechnology, biology, veterinary or related discipline; PhD degree obtained in the year of employment in the project or within 7 years before 1 January of the year of employment in the project;

  3. Experience with the following techniques: ELISA, Western blot, qPCR, and immunofluorescence staining of proteins in the cell;
  4. Experience with primary cell isolation and culture;

Post-doctoral in an NCN project SONATA BIS 9 entitled „Maternal obesity and epigenetic and metabolic regulations during gametogenesis and early embryo development in mice: the specific role of leptin signalling”.

Job Description:

The Lab of Dr Antonio Galvao is recruiting an experienced computational biologist to integrate the research programme on ovarian function in mouse models for maternal obesity. The escalating epidemic of overweight and obesity is a major burden for our health systems. Obesity leads to long-term health problems, such as diabetes, cardiovascular disease or infertility. In the lab we use mouse models and/or in vitro systems of ovarian/follicular culture to study the impact of maternal obesity in ovarian function and oocyte biology. The project investigates particularly the impact of altered leptin signalling in the ovary, a major feature of obese mothers, on the establishment of oocyte epigenome and metabolic regulations in both gamete and surrounding granulosa cells throughout folliculogenesis. The present research programme benefits from the close collaboration with the Laboratory of Dr Gavin Kelsey, Babraham Institute, Cambridge, UK, a world leader in single cell transcriptome and methylome analysis. The appointed scientist will be responsible for undertaking the analysis of methylome, transcriptome and lipidome datasets generated within the programme.

This work will lead to important advances in our understanding of the impact of altered leptin signalling in oocyte epigenome establishment, profiling also metabolic and gene expression changes in granulosa cells and other underlying mechanisms.

Key areas of responsibility:

  • quality control of NGS datasets,
  • providing high-level support in bioinformatic analysis of genome-wide epigenomic datasets,
  • integration and analysis of RNA-seq, BS-seq and other relevant datasets,
  • application of bioinformatics methods to single-cell datasets,
  • preparation of manuscripts, including sequence dataset submission to public repositories.

Person specification: Essential

  • Ph.D. in Bioinformatics or Computational biology, or Ph.D. in a Molecular biology or related discipline with a strong Bioinformatics component,
  • working knowledge of epigenomic and transcriptomic analyses,
  • programming in some of the main languages used in Bioinformatics (in particular R, Python, Java),
  • working understanding of statistics and statistical programming environments,
  • able to understand and communicate in English to a level appropriate for the position,
  • ability to work independently and as part of a team,
  • excellent organisational skills, with good time management,

Project description:

Within the OPUS grant „Milky Way to reproductive success: role of leptin in the programming of reproductive functions over generations” we aim to decipher the multiple layers of signaling pathways leading to lactocrine-based programming of reproductive performance over generations using mouse model. Because parental investment is one of the environmental conditions that can directly influence the development of offspring we decided to test how malnutrition of mother during lactation affects reproductive performance of next generations. In order to reach set specific aims of the project a combination of molecular biology, cell biology and high-throughput screening of transcriptome and epigenome will be used. We provide opportunities for interdisciplinary collaborations within the team, in the institute, as well as with the national and international partners.

Requirements:

  1. M.Sc. degree in biology, biotechnology or related area of science;
  2. Eligibility for Ph.D. studies in Poland under the supervision of prof. Kaczmarek;
  3. High motivation, independent thinking, good analytical and interpersonal skills, collaborative attitude;

Candidate qualification requirements:

  • knowledge of animal physiology and endocrinology,
  • Master’s degree in agricultural, biological, veterinary or related sciences,
  • good, documented by own research (in the form of a master’s thesis, publications from the Philadelphia list and presentation of results at scientific conferences) knowledge of the subject of animal biology and behavior,
  • experience in the application of molecular biology methods and microscopic techniques and computer analysis of results, especially ELISA, Real Time PCR,
  • research internship outside the candidate’s home country,
  • participation in research projects,
  • participation in scientific conferences,
  • very good oral and written English skills, knowledge of another foreign language will be an advantage,
  • ability to operate computer equipment,

The candidate will participate in the following research tasks:

  1. The procedure of collecting research material in the field, transporting the material to the laboratory.
  2. The isolation of oocytes from the ovaries of female roe deer, to develop a method for vitrification of oocytes.
  3. The isolation of semen from epididymides of male roe deer, develop a method for cryopreservation of semen, CASA analysis, determination of concentration and morphology.
  4. The develop in vitro fertilization method, culture embryos to blastocyst stage.
  5. Perform molecular biology techniques – Real Time PCR, electrophoresis, Western blotting, ELISA.
  6. Computer and statistical analysis of the obtained results, compilation of results for publication.

Call for a position of a PhD student in OPUS 22

Project NCN OPUS 22 entitled: „Exploration of the development, functioning and role of circadian rhythm in early life stages of Perca fluviatilis”

Principal Investigator:

Description of the position:

General aim of this project is to verify at which developmental stage the embryos or larvae of Eurasian perch, Perca fluviatilis, acquiring ‘sense of time’ and how this ability undergoes future dynamics during the larval and/or juvenile periods. Additionally, the project will allow to answer the question ‘whether’ and ‘to what extent’ modified photoperiod and/or light spectrum, affects circadian rhythm of early life stages of the species studied.

The candidate will participate in the following research tasks:

  1. Controlled reproduction of Eurasian perch (induction of ovulation and spermiation, obtaining the gametes, in vitro fertilization);
  2. Incubation of Eurasian perch eggs, evaluation of the quality of embryos;
  3. Rearing of larvae and juveniles and evaluation of their zootechnical indicators;
  4. Sampling of biological material and their evaluation in terms of transcriptomic research;
  5. RNA, protein isolations and molecular analysis;
  6. Transcriptomic and proteomic analysis;
  7. Preparation of scientific articles.

Requirements:

  1. A master’s degree in fisheries, agriculture, biology or equivalent (expected before the deadline of this application);
  2. High motivation to work in a multidisciplinary and international team with fish breeders, fish larvae as well as in molecular biology laboratory;
  3. Basic knowledge on biology and animal physiology (vertebrates);
  4. Basic knowledge on genomic tools (mainly transcriptomics and proteomics) will be appreciated, but not prerequisite;
  5. Fluency in English in writing and speaking;
  6. Availability, good work organization, ability to work in a team and deal with stress;
  7. Ability to write scientific and / or popular science papers and present the results.

Conditions of work:

  1. The selected candidate will participate in the Interdisciplinary Doctoral School of Agricultural Sciences at the Institute of Animal Reproduction and Food Research of the Polish Academy of Sciences in Olsztyn.
