Recruitment call for the NCN Preludium BIS 3 project entitled „Immunomodulatory properties of a pre-fermented whey drink enriched with chokeberry juice and colostrum” led by prof. dr hab. Barbara Wroblewska at the Institute of Animal Reproduction and Food Research of the Polish Academy of Sciences in Olsztyn, Poland.

Project description:

The PhD Student will carry out a tasks aimed at examining the effects of bioactive whey and colostrum proteins and chokeberry juice polyphenols, The first stage of the project will include determining the proportions/quantitative selection of raw materials in the mixture and determining the optimal conditions for pre-fermentation. In the next stages, simulated gastrointestinal digestion of individual components and their pre-fermented mixture will be carried out, as well as an analysis of the biological activity of the obtained hydrolysates using a model of human intestinal epithelial cells (Caco-2, cancer cells and HIEC-6, healthy cells). Epithelial cells will be cultured under standard conditions and in a state of induced inflammatory reaction (in the presence of bacterial lipopolysaccharide, LPS). The determined parameters of the immune response of epithelial cells (cell proliferation, the level of markers of the immune reaction: expression of cytokines and the level of interleukins of the pro- and anti-inflammatory pathway) will allow to unambiguously determine the immunoregulatory potential of the components and their pre-fermented mixture.

The implementation of the project will include: ongoing analysis of scientific literature, planning and performing experiments as well as laboratory and statistical analyses, interpretation of the results obtained, drawing conclusions, as well as preparing and presenting research results at scientific conferences, as well as writing scientific publications on the conducted research. Participation in the project will end with the defense of a doctoral thesis at the Institute of Animal Reproduction and Food Research of the Polish Academy of Sciences.

Department of Biodiversity Protection is the only department belonged to the Institute situated in Popielno on a peninsula, in the reserve and in the Natura 2000 area (http://popielno.pl). There are equipped laboratories in the Department: biochemical, in vitro and molecular biology. Scientific research and other tasks in which the Candidate would participate:

Post-doctoral in an NCN project SONATA BIS 9 entitled „Maternal obesity and epigenetic and metabolic regulations during gametogenesis and early embryo development in mice: the specific role of leptin signalling”.

Job Description:

The Lab of Dr Antonio Galvao is recruiting an experienced computational biologist to integrate the research programme on ovarian function in mouse models for maternal obesity. The escalating epidemic of overweight and obesity is a major burden for our health systems. Obesity leads to long-term health problems, such as diabetes, cardiovascular disease or infertility. In the lab we use mouse models and/or in vitro systems of ovarian/follicular culture to study the impact of maternal obesity in ovarian function and oocyte biology. The project investigates particularly the impact of altered leptin signalling in the ovary, a major feature of obese mothers, on the establishment of oocyte epigenome and metabolic regulations in both gamete and surrounding granulosa cells throughout folliculogenesis. The present research programme benefits from the close collaboration with the Laboratory of Dr Gavin Kelsey, Babraham Institute, Cambridge, UK, a world leader in single cell transcriptome and methylome analysis. The appointed scientist will be responsible for undertaking the analysis of methylome, transcriptome and lipidome datasets generated within the programme.

This work will lead to important advances in our understanding of the impact of altered leptin signalling in oocyte epigenome establishment, profiling also metabolic and gene expression changes in granulosa cells and other underlying mechanisms.

Key areas of responsibility:

  • quality control of NGS datasets,
  • providing high-level support in bioinformatic analysis of genome-wide epigenomic datasets,
  • integration and analysis of RNA-seq, BS-seq and other relevant datasets,
  • application of bioinformatics methods to single-cell datasets,
  • preparation of manuscripts, including sequence dataset submission to public repositories.

Person specification: Essential

  • Ph.D. in Bioinformatics or Computational biology, or Ph.D. in a Molecular biology or related discipline with a strong Bioinformatics component,
  • working knowledge of epigenomic and transcriptomic analyses,
  • programming in some of the main languages used in Bioinformatics (in particular R, Python, Java),
  • working understanding of statistics and statistical programming environments,
  • able to understand and communicate in English to a level appropriate for the position,

Project description:

Within the OPUS grant „Milky Way to reproductive success: role of leptin in the programming of reproductive functions over generations” we aim to decipher the multiple layers of signaling pathways leading to lactocrine-based programming of reproductive performance over generations using mouse model. Because parental investment is one of the environmental conditions that can directly influence the development of offspring we decided to test how malnutrition of mother during lactation affects reproductive performance of next generations. In order to reach set specific aims of the project a combination of molecular biology, cell biology and high-throughput screening of transcriptome and epigenome will be used. We provide opportunities for interdisciplinary collaborations within the team, in the institute, as well as with the national and international partners.

Requirements:

  1. M.Sc. degree in biology, biotechnology or related area of science;
  2. Eligibility for Ph.D. studies in Poland under the supervision of prof. Kaczmarek;

Department of Biodiversity Protection is the only department belonged to the Institute situated in Popielno on a peninsula, in the reserve and in the Natura 2000 area (http://popielno.pl). There are equipped laboratories in the Department: biochemical, in vitro and molecular biology. Such a location provides isolation from the city noise, contact with nature, but at the same time, access to the nearest city (Ruciane-Nida), located about 20 km away, does not take more than 15-20 minutes.

Scientific research and other tasks in which the Candidate would participate:

  • research related to the genetic diversity of free-living animals using transcriptomic and proteomic methods,

  • searching for breeding markers used in the development of effective techniques to reduce or intensify reproductive processes,
  • studies on the dependence of microbiota on immunological parameters of the organism of free-living animals,
  • development of non-invasive hormonal monitoring in relation to reproduction, behavior, etc. in free-living animals.
  • influence of environmental factors, such as feeding base, antroporession on homeostasis of selected animal species,
  • analyses in the laboratory,
  • work with live domestic animals and collection of research material from domestic and free-living animals, vividly and post mortem.

Candidate qualification requirements:

  • knowledge of animal physiology and endocrinology,
  • Master’s degree in agricultural, biological, veterinary or related sciences,
  • good, documented by own research (in the form of a master’s thesis, publications from the Philadelphia list and presentation of results at scientific conferences) knowledge of the subject of animal biology and behavior,
  • experience in the application of molecular biology methods and microscopic techniques and computer analysis of results, especially ELISA, Real Time PCR,
  • research internship outside the candidate’s home country,
  • participation in research projects,
  • participation in scientific conferences,
  • very good oral and written English skills, knowledge of another foreign language will be an advantage,
  • ability to operate computer equipment,
  • practical ability to drive a passenger car (category B, at least 2 years),
